摘要
目的研究小鼠胚肺组织感染小鼠巨细胞病毒(murinecytomegalovirus,MCMV)后其细胞热休克蛋白70(HSP70)的表达情况,为临床病毒感染性疾病的诊断开辟新途径。方法体外培养小鼠胚肺细胞,加入MCMV(感染组)后4、8、12、16、24、48、72、96小时分别收获细胞;用免疫组织化学方法检测HSP70表达情况并与对照组(不感染病毒)比较。结果感染组各时相HSP70表达明显增强,其中96小时表达最强,8小时次之,24小时最弱;随HSP70表达增强细胞变性程度加重。结论MCMV可诱导小鼠胚肺细胞HSP70的高表达;HSP70表达强度与细胞病变程度呈正相关。
Objective To study the expression of heat shock protein 70 (HSP70) of the murine embryo lung cells infected by murine cytomegalovirus (MCMV) in vitro and looking for a new approach to diagnose clinical virus infective disease.Methods The murine embryo lung cells were cultivated in vitro and harvested after MCMV infection (infective group)4?8?12?16?24?48?72?96 hours respectively.The expression of HSP70 was detected by immunohistochemistry and compared with control group.Results Expression of HSP70 enhanced obviously at each time in infective group compared with the control group,the strongest at 96 hour,the second at 8hour and the weakest at 24hour.The degree of cellular degeneration aggravated with enhancing expression of HSP70.Conclusions MCMV can induce the high expression of HSP70 which correlated with the degree of cellular metastasis.
