摘要
目的探讨结核病与膜白介素-2受体(mIL-2R)、可溶性白介素-2受体(sIL-2R)表达水平的关系。方法采用生物素-链霉亲和素(Biotin-Streptavidin,BSA)系统及双抗体夹心ELISA法检测结核病患者mIL-ZR及sIL-2R的表达水平。结果结核病患者PHA诱导前后 mIL-2R水平分别为(3.28±1.35)%和(29.18±5.86)%,与对照组相比,差异有显著性(P<0.01和P<0.05);结核病患者血清中sIL-2R水平为(474.3±102.5)%,与对照相比,差异有显著性(P<0.01)。有空洞型肺结核的mIL-2R、sIL-2R表达水平的变化较无空洞者更为明显(P<0.01)。无空洞型肺结核,与结核性胸膜炎、肾结核的mIL-2R、sIL-2R表达水平相似,差异无显著性(P>0.05)。结论结核病患者体内mIL-2R表达水平降低,sIL-2R水平升高,结核病患者体内存在明显细胞免疫功能低下,其外周血单个核细胞(PBMC)对PHA的诱导反应能力降低。结核的病变部位、病变程度及预后与患者的mIL-2R、sIL-2R表达水平有一定关系。
Objective To explore the relationship between tuberculosis and expression of membrane in- terleukin-2 receptor (mlL-2R) as well as the level of soluble interleukin-2 receptor. Methods The expression of mlL-2R and the level of slL-2R were detected by BSA and sandwiched ELISA. Results Before and after in- ducement of phytohemagglutinin (PHA), the levels of mlL-2R of the patients with tuberculosis were (3.28± 1.35)% and (29.18±5.86)% respectively. There was significant difference between patients and normal controls (P<0.01 and P<0.05). And the levels of slL-2R in sera of patients with tuberculosis were (474.3±102.5)%, It was significantly higher in patients than that in normal controls (P<0.01). The expressive level of mlL-2R and slL-2R of pulmonary tuberculosis with cavitation was more obvious than that without it (P<0.01). The expres- sive level of mlL-2R and slL-2R in pulmonary tuberculosis with no cavitation was similar to tuberculous pleurisy and kidney tuberculosis and there was no significantly difference (P>0.05). Conclusion The expressive level of mlL-2R and sKIL-2R in patients with tuberculosis changed and the immunological function of body was in disorder. The function of cellular immunity and response to PHA were far lower obviously in tuberculosis than in normal controls. The focus of tuberculosis, the pathological changes and prognosis were associated with the level of mlL-2R.
