摘要 目的比较逆转录聚合酶链反应法(RT-PCR)与血凝抑制法在流感病毒鉴定中的优劣,以期建立能快速、特异地从临床标本中检测并鉴定流感病毒的新方法。方法用常规细胞培养法增殖病毒后,分别用RT-PCR反应法和常规血凝抑制法进行流感病毒鉴定。结果在13份发生细胞病理变化的标本中RT-PCR阳性标本为9份,其中甲型7份(6份甲3亚型、l份甲l亚型),乙型2份;而用血凝抑制法鉴定阳性标本仅6份。结论RT-PCR法快速、特异、灵敏,在流感病毒鉴定中具有较大的应用价值。 Objective To compare the advantage and disadvantage of RT-PCR assay and HI test in the detection of Influenza virus, so as to establish proper method for differentiation of influenza virus from clinical samples. Methods After amplifing the virus by cell culture, influenza virus was differentiated by using RT-PCR and hemagglutin ation inhibition test. Results Out of the 13 samples which had cytopathologic changes, 9 positive strains were detected by RT-PCR assay including seven A type (six H3N2 subtypes and one H1Nlsubtype) and two B types, but 6 positive strains were detected by HI test. Conclusion RT-PCR is rapid, specific and sensitive and possesses great value for practical application in the detection of influenza virus.
