摘要
本文采用薄层扫描及盐酸沉淀蛋白法系统地考察了从兔肝匀浆中提取士的宁生物碱的条件。实验设计按正交法进行,选用L_(16)^((2)15)正交表。考察了盐酸浓度、水浴温度、加热时间及溶液碱化PH 值等主要因素。结果表明:盐酸浓度为10%,80℃水浴加热15分钟,碱化PH 为12,在此条件下,从兔肝匀浆中提取士的宁生物碱回收率可达60.03%。操作简便,省时,优于目前较常用的几种方法。实验结果证实:在所选条件下,对硫酸奎宁及硫酸奎尼丁等生物碱,也可得到较高提取回收率。提示本法有推广应用的前景。
The extraction conditions of strychninefrom rabbit liver homogenate by using hyd-rochloric acid to precipitate proteins havebeen systematically investigated.The exper-iment was designed by using L16^((2)15) or-thogonal table and TLC—densitometry,which was used to determine the extractionrecovery.The optimization conditions are asfollwing;concentration of HCL 10%,temp-erature of water-bath 80℃,time of heating15 minutes and the PH value of tha extra-cted solution 12.In these conditions,stryc-hnine could be easily extracted from rabbitliver homogenate,with a recovery over60.03%.Other alkaloids such as Quinine andQuinidine also could be extracted from rabbitliver hodogenate by using this method.
