颅内动脉瘤细胞外基质相关基因表达谱的基因芯片研究 被引量:13

摘要 目的检测颅内动脉瘤中细胞外基质相关基因的表达谱,探讨其在颅内动脉瘤发病机制中的作用。方法利用Affymetrix公司的人类基因组基因芯片U133A分别检测3例颅内动脉瘤组织和3例正常颅内动脉组织中的基因表达谱,用生物信息学软件进行比较分析,对差异表达基因进行功能分类,对其中的细胞外基质相关基因进行分析。结果与正常颅内动脉组织相比,颅内动脉瘤中有383个基因的差异表达水平达10倍以上;在这383个基因中,有23个基因与细胞外基质有关,其中11个基因的表达水平上调,12个基因的表达水平下调。结论细胞外基质相关基因的异常表达可能参与了颅内动脉瘤的形成和发展。 Objective To examine the gene expression profile related with extracellular matrix in intracranial aneurysm,and to analyze the role of these genes for the pathogenesis of intracranial aneurysm.[WT5”HZ]Methods Three intracranial aneurysms tissuses removed during the operation and 3 normal intracranial arteries from 3 normal adults were collected.The cDNA was retrotranscribed from the extracted RNA and the biotin-labeled cRNA derived from the transcription of cDNA was fragmented as probes,respectively.The probes were hybridized with Affymetrix Human Genome U133A Array.GeneArray Scanner was used to screen the signals of hybridization and Microarray Suite software 5.0 was applied to detected the expression of genes.Genes related with extracellular matrix were analysized.Results 383 genes were showed to be differently expressed more than 10-fold between normal intracranial artery and intracranial artery.Among them,23 genes related with extracellular matrix.11 genes were overexpressed and 12 genes underexpressed in intracranial aneurysm.Conclusion Genes related with extracellular matrix differently expressed in intracranial aneurysm maybe involved in the pathogenesis of intracranial aneurysm.
