摘要
目的:构建人牙囊细胞的体外三维立体培养模型,探讨两种支架材料用于牙周组织工程的可行性。方法:实验于2003-01/2004-10在第四军医大学口腔医学院组织工程实验中心完成。采用酶消化法原代培养人牙囊细胞,传代扩增后,取第4代细胞分别接种于煅烧骨和Ⅰ型胶原膜三维支架上,体外继续培养1周,进行细胞记数和扫描电镜观察细胞生长状况。结果:细胞在两种材料上均能良好贴附并增殖,扫描电镜可见人牙囊细胞在这两种支架材料上均黏附良好,增殖旺盛,伸展充分,并有基质分泌。结论:煅烧骨和冻干胶原膜均有望成为牙周组织工程支架材料。
AIM:To construct a three-dimensional culture model of human dental follicle cells(DFCs) to evaluate the feasibility of the two materials in the scaffolds of periodontal tissue engineering. METHODS:The experiment was conducted in the Department of Dentistry,Nanfang Hospital,Southern Medical University from January 2003 to October 2004.Human DFCs were cultured by enzyme digestion for proliferation and passage.Cells of the fourth passage were seeded on a three-dimensional sintered bone and lyophilized type Ⅰcollagen membrane to subculture in vitro for another 1 week.The cells were counted and their growth on the scaffolds was observed by scanning electron microscope(SEM). RESULTS:DFCs adhered well and proliferated on the two three-dimensional scaffolds.SEM results showed the DFCs adhered eugenically,proliferated adequately and extended excessively on the scaffolds accompanied by secretion of matrix. CONCLUSION:Both of sintered bone and lyophilized collagen membrane are expected to be the materials for scaffolds in periodontal tissue engineering.
