猫儿眼提取物对荷瘤小鼠肝癌细胞毒性及免疫功能的影响 被引量:8

摘要 目的:探讨猫儿眼提取物对荷瘤小鼠肝癌细胞的毒性作用和对免疫功能的影响,并以5-氟尿嘧啶为标准对照。方法:实验于2003-07/10在兰州军区总医院实验动物科完成。选BALB/c小鼠120只,于皮下接种肝癌瘤液0.01mL/g体质量。随机分为4组,每组30只。接种肝癌瘤液24h后,模型组给予等量蒸馏水;5-氟尿嘧啶组每天腹腔注射5-氟尿嘧啶100mg/kg体质量;甘肃猫儿眼提取物1组每天口服甘肃猫儿眼提取物10g/kg体质量;甘肃猫儿眼提取物2组每天口服甘肃猫儿眼提取物20g/kg体质量。连续给药10d后,分别取40只(每组取10只)对荷瘤小鼠瘤体、胸腺和脾脏质量以及免疫功能的影响进行测定分析。结果:120只小鼠均进入结果分析。①甘肃猫儿眼提取物对肝癌细胞增殖的抑制作用:5-氟尿嘧啶组和甘肃猫儿眼提取物2组肝癌细胞的抑制率分别为24.6%和64.7%;甘肃猫儿眼提取物1组肝癌瘤质量比对照组增长了4.7%。②甘肃猫儿眼提取物对肿瘤组织的毒性作用:对照组肝癌细胞生长活跃,核分裂像较多见;5-氟尿嘧啶组、给药组肝癌瘤体均见部分坏死,或在坏死灶边缘出现纤维组织增生,间质水肿。③甘肃猫儿眼提取物对肝癌荷瘤小鼠免疫功能的影响:与对照组比,甘肃猫儿眼提取物2组小鼠碳粒廓清指数提高了59.4%,胸腺和脾脏的质量分别增加了51.9%和33.0%,血清溶血素值增高了31.2%。结论:猫儿眼提取物比5-氟尿嘧啶对肝癌细胞增殖具有更明显的抑制作用,且剂量高抑制作用强,其作用与提高荷瘤小鼠免疫功能有关。 AIM:To study the cytotoxic effect of maoeryan extract on liver cancer strain cells and its effect on immune function in tumor-bearing mice,and compare with the effect of 5-fliorouracil as the standard control. METHODS:The experiment was completed in the Department for Experimental Animals,General Hospital of Lanzhou Military Area Command from July to October 2003.After 120 healthy BALB/c strain mice were subcutaneously injected with liver cancer strain cells(0.01 mL/g),they were randomly divided into 4 groups with 30 mice in each. Equivalent distilled water(100 mL) was given in the model group.Mice in 5-fliorouracil group was injected with 5-fliorouracil 100 mg/kg intraabdominally per day,those in Gansu maoeryan extract group 1 took maoeryan extract 10 g/kg orally per day,and those in the Gansu maoeryan extract group 2 took maoeryan extract 20 g/kg orally per day.After 10 consecutive days' administration,10 mice of each group were chosen for the detection and analysis of effect of maoeryan extract on the mass of tumor,thymus gland and spleen,and the immune function. RESULTS:All the 120 mice were involved in the result analysis.①Inhibitory effect of maoeryan extract on the proliferation of liver cancer cells:The inhibition rate was 24.6%and 64.7%in the 5-fliorouracil group and Gansu maoeryan extract group 2 respectively,the mass of tumor in liver cancer was increased by 4.7%in the Gansu maoeryan extract group 1 as compared with the model group.②Cytotoxic effect of maoeryan extract on the tumor tissue:The growth of liver cancer strain cells was active,and karyokinesis was found mostly in the model group.Partial necrosis of liver cancer tumor or hyperplasia of fibrous tissues around focal necrotic tumor with interstitial edema was found in the 5-fliorouracil group and Gansu maoeryan extract groups.③Effect of maoeryan extract on the immune function in the tumor-bearing mice:The clearance index was increased by 59.4%,mass of thymus gland and spleen by 51.9%and 33.0%,and complement hemolytic activity(CH50) by 31.2%in the Gansu maoeryan extract group 1 as compared with the model group. CONCLUSION:The maoeryan extract has inhibitory effect on the proliferation of liver cancer cells more obviously than 5-fliorouracil has.The higher dose of maoeryan extract is, the more obvious is its inhibitory effect.Maoeryan extract participates in the enhancement of immune function in tumor-bearing mice.
