摘要
目的观察加味桃核承气汤及不同提取物对糖尿病大鼠心肌纤维化的影响。方法用链脲佐菌素腹腔注射复制糖尿病大鼠模型,观察加味桃核承气汤全方煎剂及水提醇沉、正丁醇、乙酸乙脂不同提取处理成分群对糖尿病大鼠心肌纤维化影响,并与达美康对照,共治疗8周。测定心脏湿重/体重比、心肌羟脯氨酸及Ⅰ、Ⅲ型胶原mRNA表达。结果各组心脏/体重比值与空白组比较,全方组未显示出差异,其余各组均显著增加(P<0.01);心肌羟脯氨酸含量测定也显示出相似的效果;各组心肌Ⅰ型胶原mRNA表达均较空白组下降,但无统计学意义;Ⅲ型胶原蛋白mRNA表达模型组较空白组显著升高,中药全方、水提醇沉及正丁醇组较模型组显著降低(P<0.05-0.01)。结论加味桃核承气汤全方煎剂及水提醇沉、正丁醇提取物能阻止糖尿病大鼠心肌胶原异常增生,维持Ⅰ/Ⅲ型胶原正常比值,作用似优于达美康。
OBJECTIVE To observe the effect of Taohe Chengqi Decoction and different extractions on myocardial fibrosis of rats with diabetes. METHOD Diabetes rat models were made using streptozotocin, and observation was made on the effect of the decoction and different extractions on myocardial fibrosis of the rats with diabetes, compared with a group treated with Diamicron. The treatment lasted for 8 weeks. RESULT Compared with the blank group, the heart/body weight ratio in the prescription group did not show any marked difference, but this ratio rose markedly in all the other groups (P < 0.01) . Similar results were obtained in the hydroxypro line content determination. The mRNA expression of type Ⅰ myocardium collagen in each group dropped, but with no statistical significance. The mRNA expression of type Ⅲ myocardium collagen rose markedly in the model group compared with the blank group, but dropped in the prescription group and extraction groups (P<0.05, P < 0.01). CONCLUSION Taohe Chengqi Decoction and its extractions can prevent the abnormal hyperplasia of myocardium collagen of rats with diabetes and maintain the normal Ⅰ/Ⅲ collagen ratio, with better functions than Diamicron
