摘要
采用RAPD分子标记技术对广西何首乌野生种质资源进行遗传多样性分析。结果表明:40对随机引物用于PCR扩增,共得到295条扩增DNA片段,175个多态性片段,多态性达到59.3%,其中田林种源与西林种源之间的遗传变异最小,西林种源和灌阳种源之间遗传变异最大;根据RAPD标记划分10个何首乌野生种源得到5个品种群,这5个类型与地理分布有一定关系,地理位置较近的材料能聚在一起。分子标记可用于何首乌种质资源的分类、真伪鉴定及良种选育。
The ten wild germplasm resources of Guangxi were collected and their inheritable variety were analyzed by the RAPD.The result showed that the PCR amplification with 40 screened primers were conducted to access and 295 genes were obtained from the PCR Amplification,175(59.3% of total genes) of which were polymorphic.The genetic variation is lowest between the germplasm resource of Tianlin and the germplasm resource of Xilin,and the genetic variation is biggest between the the germplasm resource of Xilin and the germplasm resource of Guanyang.Five tapes of ten Polygonum multilorum classified by RAPD were related with their distribution,and some of them from near distribution can get together.The result by the RAPD would offer to scientific theories for the classification,authenticity and the good breeds selective of the germplasm resources of the P.multilorum.
