斑叶海棠薄层培养中细胞无丝分裂及2,4-D的特殊效应的研究 被引量:7

摘要 以经过预培养的斑叶海棠为材料,取叶柄表层3—4层细胞,切成8×10mm大小作为外植体,所用培养基采用MS基本成份附加不伺浓度的2,4-D。结果如下:1.在含0.1mg/l 2,4-D的培养基上,大多数叶柄表皮细胞可以进行一次无丝分裂。与有丝分裂一样,无丝分裂中有核仁和核膜的消失和再现。分裂过程中核始终靠近细胞壁,细胞分隔部分的伸展也是单向的。2.实验中2,4-D对表皮细胞产生了很特殊的效应。在0.1mg/l 2,4-D的培养基上培养45小时后,细胞都发生收缩。收缩以前大多数细胞都有过一次无丝分裂。如果2,4—D的浓度降至0.01mg/l,细胞将反复进行无丝分裂形成疏松的愈伤组织而不发生收缩。 Explants were obstained by excising cortical tissues from petioles of precultured Begonia rex-m(?)torum in 3×10mm strips, containing 3-4 cell layers. The medium used was basic MS supplemented with 2,4-D in different concentrations. Results obtained were as follows: 1. Most epidermal cells of petioles could divide once in amitosis on MS medium supplemented with 0.1mg/1 2,4-D. Disappearance and reformation of nucleoli and nuclear membranes could be seen in amitosis as in mitosis. Nuclei kept close to the wall in all the procedure, and cell plate extended in only one direction. 2. 2, 4-D had a special effect in thin cell layer culture of Begonia rex petioles. It was interesting that protoplasm of epidermal cell shrank after 45-hour culture on MS added with 0.1mg/1 2,4-D. Most cells had divided once before the shrank occured. If the concentration of 2, 4-D was decreased to 0.01mg/1, cells could divide in amitosis repeatedly, and form loose callus. And no cell shrink occured.
