培养贮脂细胞运动收缩功能的研究

摘要 用胶原酶灌流法及泛影葡胺比重离心法分离大鼠贮脂细胞,培养的细胞抗Dismin抗体间接荧光染色阳性。经显微录像装置摄影观察及画像解析装置(Luzex500)测定,可见其能游走样移动,分裂时突起收缩,分裂后又伸展。添加血管收缩物质能诱发贮脂细胞收缩,除去该类物质后,收缩的细胞又渐伸展,表明贮脂细胞有运动收缩功能。 The wister fat-storing cells were isolated by perfusion with collagenase and centrifugation with metrizamide density cushion techniques and cultured in vitro.The fat storing cells were confirmed by the presetnlation of the lipid drop in the cytoplasma and the visualization of Dismin with anti-Dismin antibody by using indirect immunmofluorescence method.By use of the videotape recorder (VIR) and the imagine analysis system (Luzex 500),we observed the wandering immigration,the abrupt contraction of fat-storing cells with spike,and their change into ball like shape in its fission phase.After that the cell began to stretch.The contraction of this cell can be induced by the presence of endothelin-l (Et-1) 10-2 mmol/L,substance P (SP) 1mmol/L and Noradrenaline 2×10^(-5) mmol/L.The con tracted cells become stretched when treated with the ET-1 and SP but nor-adrenaline.All these results indicate that the fat-storing cells have function of contraction and movement.
