摘要
目的:探讨四逆汤类方提取物对离体培养的乳鼠心肌细胞缺氧/复氧损伤的保护作用。方法:培养的乳鼠心肌细胞经终浓度分别为1、5、25μg/mL的四逆汤类方提取物预处理,进行缺氧/复氧损伤后,比色法测定细胞内线粒体脱氢酶活性、丙二醛(MDA)含量及培养液上清中乳酸脱氢酶(LDH)活性。结果:与对照组比较,附子、干姜配葱白提取物(EAGO),附子、干姜配甘草提取物(EAGL),附子、干姜配胆汁提取物(EAGB),附子、干姜配人参提取物(EAGG)可以提高细胞内线粒体脱氢酶活性,与附子配干姜提取物(EAG)组比较无显著差异。EAGO、EAGL、EAGB、EAGG可以降低上清液中乳酸脱氢酶活性,与EAG组比较差异显著。EAGL、EAGG可以降低细胞内MDA含量,与FAG组比较差异显著。结论:四逆汤类方均可以提高缺氧/复氧损伤的乳鼠心肌细胞内线粒体脱氢酶活性,降低上清液中乳酸脱氢酶活性。四逆汤和人参四逆汤提取物有降低细胞内MDA的作用。提示四逆汤类方对缺氧/复氧损伤的心肌细胞均有保护作用。
Objective:To study the protective effect of the extracts from Sini Tang Serial Decoctions (ESTSD)against hypoxia/reoxygenation(H/R) injury on neonatal rat cardiomyocytes.Methods:Cultured neonatal rat cardiomyocytes were preconditioned with ESTSD in different concentrations(1,5,25 μg/mL)respectively.Then after H/R treatment the enzymatic activity of intracellular mitochondria dehydrogenase and the contents of intracellular maleic dialdehyde(MDA)as well as the activity of extracellular lactate dehydrogenase(LDH)were measured using colorimetric method.Results:Compared with the control group,the ESTSD including extracts from Aconite root,Dried ginger and Fistular onion stalk(EAGO),extracts from Aconite root,Dried ginger and Prepared licorice root(EAGL),extracts from Aconite root,Dried ginger and pig's Bile(EAGB)and extracts from Aconite root,Dried ginger and Ginseng(EAGG)could increase significantly the activities of intracellular mitochondria dehydrogenase and had no significant difference compared with extracts from Aconite root and Dried ginger (EAG).EAGO,EAGL,EAGB,EAGG all could significantly decrease the activities of extracellular LDH compared with the group and EAG.EAGL and EAGG decreased significantly the contents of intracellular MDA compared with the control group and EAG.Conclusion:The results showed ESTSD increased significantly the activities of intracellular mitochondria dehydrogenase,but decreased significantly the activities of extracellular LDH.EAGL and EAGG decreased significantly the contents of intracellular MDA.ESTSD has a protective effect against H/R injury on cultured neonaltal rat cardiomyocytes.
