摘要
目的研究摄入不同剂量碘化钾后胸腺细胞角蛋白19阳性(CK19+)上皮细胞的变化。方法取断乳后1个月的Wistar大鼠,随机分为6组:低碘组、适碘组、5倍碘组、10倍碘组、50倍碘组、100倍碘组,分别给予不同浓度的碘化钾。饲养6个月后,测定血清甲状腺激素水平,胸腺上皮细胞免疫组化染色。结果与适碘组相比,100倍碘组和低碘组的T4、T3和rT3均显著降低(P<0.05),并且可见这两组的胸腺上皮细胞局限性增生;50倍碘组、100倍碘组和低碘组胸腺CK19+上皮细胞的体密度和面数密度显著降低(P<0.01);其余各组未见明显异常。结论长时间的摄碘异常(碘缺乏、碘过量)可使大鼠甲状腺激素水平降低,从而导致胸腺CK19+上皮细胞减少,胸腺退化。
Objective To study the changes of CK19+ thymic epithelial cells (TEC) after intake of differentconcentrations ofPotassiumiodide(KI). Methods Wistar rats, 1 month after stoppinglactation, were divided randomlyinto 6 groups by giving various concentrations of KI: low iodine(LI), normal iodine(NI), 5 folds of iodine (5HI),10 folds of iodine(10HI), 50 folds of iodine(50HI) and 100 folds iodine(100HI). Thyroid hormones in serum weremeasured, and then immunohistochemical staining of thymic epithelial cells were performed 6 months later. ResultsT4、T3 and rT3 decreased dramatically compared with those of normal group(P < 0.05), local hyperplasia of thymicepithelial cells(TEC) was seen in 100HI as well as in LI groups. Volume density (Vv) and numerical density inarea(NA) of CK19+ reduced significantly in 50HI, 100HI and LI groups(P < 0.01). Conclusions Long time ofintake of overdosed KI or iodine deficiency results in decreased thyroid hormone level, thereafter thymusdegeneration and decrease of CK19+ thymic epithelial cells.
