地方性氟中毒患者外周血淋巴细胞细胞周期的实验研究

摘要 目的运用流式细胞术研究氟对人体外周血淋巴细胞细胞周期分布的影响。方法从病情程度不同的氟中毒患者和正常对照者外周血中分离淋巴细胞,用缓冲液和溶血素清洗和去除杂质后,调整细胞数约3×1012细胞/L,PI染色使细胞核发射出荧光,用FACSCaliburTM流式细胞分析仪检测淋巴细胞细胞周期分布、DNA相对含量和细胞增殖力。结果氟中毒患者与正常对照者相比,外周血淋巴细胞细胞周期分布差异无统计学意义(FG0/G1=0.84,P=0.4755;HS=4.06,P=0.2550;HG2/M=4.47,P=0.1881),并且淋巴细胞增殖指数和DNA相对含量差异亦无统计学意义(FPI=0.8369,P=0.4776;FDNA=0.3867,P=0.7629)。结论地方性氟中毒患者体液中的含氟量,尚不足以引起外周血淋巴细胞细胞周期的改变。提示氟对细胞周期的影响可能既存在周期依赖性,又有种属特异性以及剂量效应关系存在。 Objective To study the effect of fluorine on distribution of cell cycle of peripheral bloodlymphocyte of patients with endemic fluorosis by flow cytometry (FCM). Methods Lymphocytes isolated fromperipheral blood of patients with fluorosis at different degrees and the normal controls were adjusted to a cellconcentration of 3 ×1012/L after it was washed by buffer solution and hemolysin and then impurities was removed.For nucleus emitted fluorescence after PI dyeing, cell cycle distribution of lymphocytes, relative contents of DNAand proliferation ability of cells could be detected with FACSCaliburTM FCM analyzer. Results The detection ofcell cycle showed that there were no obvious differences in distribution of cell cycle of peripheral blood lymphocyte(FG0/G1 = 0.84, P = 0.475 5; HS = 4.06, P = 0.255 0; HG2/M = 4.47, P = 0.188 1) , relative contents of DNA andproliferation ability of cells (FPI = 0.836 9, P = 0.477 6; FDNA = 0.386 7, P = 0.762 9) between the patients withfluorosis and the normal controls. Conclusions The concentration of humoral fluorine in patients with endemicfluorosis inadequately results in the change of cell cycle of peripheral blood lymphocyte. It indicates that cycledependence and species specificity as well as dose-effect relationship exist in the influence of fluorine on cell cycle.
