摘要
目的研究脑出血后延髓内脏带(MVZ)内FOS蛋白表达变化规律,探讨MVZ在脑出血致多器官功能障碍综合征(MODS)时的中枢调控机制。方法①采用0.8 U胶原酶注入大鼠尾状核构建脑出血模型,54只Wistar 大鼠随机被分为正常对照组,假手术组,出血后4 h、8 h、12 h、24 h、36 h、48 h和72 h时相点的7个亚组;②应用免疫组织化学ABC法观察各时相点MVZ内FOS蛋白的表达。结果对照组和假手术组MVZ内FOS蛋白阳性表达极少;出血组FOS蛋白阳性细胞主要分布于MVZ的弧形带状区内,在孤束核、腹外侧网状核和迷走神经背核均见密集表达,24-36 h达高峰,在72 h仍有阳性表达。结论MVZ参与了对脑出血后周围器官功能的调控,可能是脑出血致MODS的直接调控中枢之一。
Objective To investigate the changes of FOS protein expression in the medullary visceral zone(MVZ) of the rat following cereral hemorrhage; to discuss the possible center regulation mechanism of MVZ in the rat following cereral hemorrhage by multiple organ dysfunction syndrome(MODS). Methods (1)Cerebral hemorrhage was induced in rats by injecting 0.8 U collage-nase Ⅶ into the caudase putamen. 54 Wistar rats were randomly divided into 9 groups: normal group, sham-operative group and hemorrhage groups including 4 h、8 h、12 h、24 h、36 h、48 h、72 h seven time points. (2)The area density of positive staining expressing FOS protein were analyzed for the relative content of immunohistochemistry( ABC). Results A few of FOS protein expressed in the MVZ of normal control group. The FOS-like neurons were densely expressed in the MVZ of cerebral hemorrhage group, especially within the nucleus of the solitary tract、dorsal nucleus of vagus nerve and ventrolateral medulla. FOS protein expressed in the MVZ with time progress reaching the peak at the 24 - 36 h,FOS-like neurons can be observed at the 72 h still. Conclusions MVZ may contribute to dysfunction of peripheral organs. It may be one of the regulation center after cerebral hemorrhage by MODS.
