摘要
为探求AT5BIVA细胞辐射敏感性与超氧阴离子自由基O2.-之间的联系,以正常人皮肤成纤维细胞GM0639为对照,用3HTdR掺入法和细胞色素C还原法分别测定正常及照射条件下细胞增殖能力及其培养介质中O2.-浓度(nmol/106细胞)。结果表明,在正常培养条件下,两种细胞的增殖能力和向培养介质中释放O2.-的量均随培养时间的延长而增加。在不同剂量60Coγ射线照射条件下,两种细胞的增殖能力和向培养介质中释放O2.—的量均随剂量增大而减小;两种细胞的增殖能力均与其培养介质中O2.—浓度密切相关,且AT5BIVA细胞的增殖速率大于GM0639细胞。提示AT5BIVA细胞高辐射敏感性与O2.—产生量密切相关。
To study the correlation between the radiosensitivity of AT5BIVA fibroblast and superoxide anion free radical O_2 .- ,and after exposure to 60 Co γ ray,comparison was made with GM0639 fibroblast 3 H-TdR incorporation method and cytochrome C reduction method were used to determine cell proliferation(CPM)and the concentration(nmol/h/10 6 cells) of O_2 .- in culture medium under normal culture condition.The results showed that both the proliferation rate of two kinds of fibroblasts and the release of O_2 .- into the medium increase with extended time under normal culture condition,but decrease with extended time under different 60 Co γ ray exposure conditions.The proliferation rate of two kinds of fibroblasts are closely related to the concentration of O_2 .- in the medium.AT5BIVA fibroblasts proliferates at a higher rate than GM0639 fibroblasts do.The findings suggest that the high radiosensitivity of AT5BIVA fibroblast closely correlates with its O_2 .- production.
