The Study of Growth Factors in Human Colostrum

摘要 应用 ̄3H胸腺嘧啶掺入技术及一系列生化、分离鉴定手段,分析测定了人初乳中生长活性因子的组成与性质。结果:(1)初乳中含有丰富的生长因子,可促进体外培养的NIH-3T_3细胞的DNA合成,且培养液在含0.5%(V/V)初乳时对细胞增殖的促进作用与含3.0%小牛血清相同。初乳每毫克蛋白促细胞DNA合成的作用比牛血清高20倍。(2)人初乳、成熟乳和牛初乳的生长活性因子比较,人初乳中生长活性因子最为丰富,含有两类生长活性因子,分别为初乳酸性生长因子(CAGF)和碱性生长因子(CBGF)。(3)上述两种因子对尿素和盐酸具稳定性,其中CAGF不能被巯基乙醇灭活,而CBGF可被巯基乙醇灭活。提示:CAGF为表皮生长因子样因子;CBGF为血小板源生长因子样因子。这两类生长活性因子均可促进和维持细胞的正常增殖与分化。 bjective To characterize and identify the growth factors in human colostrum and to evaluate theimportance of breast-feeding.Methods The activity of growth factors in human colostrum was deterrnined by technique of  ̄3H-TdR incorporation into cuItured NIH- ̄3T_3 cells. The acid growth factor (CAGF)and basic growth factor ( CBGF ) were purified from human colostrurn by a sequence of chromatography. The study of stability and SDS-PAGE was applied to identify the CAGF and CBGF. ResultS0.5%(v/v)of human colostrum and3.0 %(v/v)of bovine serum bad the same activity in stimulating DNA synthesis. The specific activity of human colostrum in stimulating DNA synthesis was 20 times greater than that of bovine serum,The activity of growth factors in human colostrum was higher than that in human milk or bovine colostrum,and only human colostrum contained two different kinds of growth factors-CAGF and CBGF Coucl usions Human colostrum contains two kinds of growth factors.CAGF is epidermal growth factor like(EGF-like)growth factor and the CBGF is platele differentiation growth factor like(PDGF-like)growth factor.The effects of human colostrunm on promoting baby growth and development is stronger than that of human milk and bovine colostrurn.
