针灸对克罗恩病大鼠结肠胶原合成及TGFβ mRNA表达的调节 被引量:3

摘要 目的:观察针灸对克罗恩病大鼠结肠Ⅰ、Ⅲ、Ⅳ型胶原蛋白合成及转化生成因子-β(TGFβ)mRNA表达的影响。方法:在建立克罗恩病大鼠模型基础上,进行针灸治疗。采用免疫组织化学方法检测各组大鼠结肠黏膜Ⅰ、Ⅲ、Ⅳ型胶原表达;用RT-PCR方法检测各组大鼠脾脏、结肠组织TGFβmRNA表达情况。结果:与正常组大鼠比较,模型组大鼠结肠Ⅰ、Ⅲ、Ⅳ型胶原及脾脏、结肠TGFβmRNA表达均异常升高,隔药灸治疗可明显抑制克罗恩病大鼠纤维化进程,降低结肠Ⅰ、Ⅲ、Ⅳ型胶原及脾脏、结肠TGFβmRNA表达。结论:结肠组织纤维化是克罗恩病大鼠的主要病理表现之一,隔药灸治疗大鼠克罗恩病肠纤维化的可能机制是通过下调克罗恩病大鼠结肠TGFβmRNA过度表达,从而阻断或抑制细胞外基质(Ⅰ、Ⅲ、Ⅳ型胶原)的合成,促使其组织结构与功能改善。 Objective: To observe the effect of Acupuncture and Moxibustion on ,, collagens and TGFβmRNA in colon of Crohns disease(CD) rats. Method: The CD rats model was established by TNBS clusis. When the model established successfully, SD rats were randomly divided into normal group, CD model group, herbs-partition moxibustion(HPM) group and electro-acupuncture(EA) group. The HPM group and EA group rats were treated by herbs-partition moxibustion and electro-acupuncture respectively.,, collagens expression in colon were detected by immunohistochemistry and TGFβ mRNA expression in colon and in spleen were detected by RT-PCR. Result: Compared with the normal rats, the expression of , , collagen and TGFβ mRNA were abnormality increased in model rats, and were markedly down-regulated by herbs-partition moxibustion. Fibrosis of colon in CD rat was also restrained by the treatment. Conclusion: These data suggest that fibrosis is one of the pathological changes in CD rat. Herbs-partition moxibustion treatment may be therapeutically beneficial in colon fibrosis of CD rats by down-regulated TGFβ mRNA expression in colon tissues to block or suppress the ,, collagens synthesis, then melioration the structure and the function of colon.
