摘要
运用 ̄(125)I-EGF放射受体法,探讨对不同周龄Wistar鼠使用雄、雌激素过程中及停药后睾丸表皮生长因子(EGF)与EGF受体结合情况。结果表明:EGF与睾丸细胞膜在27℃孵育70分钟结合最大,年龄增长与睾丸重量呈正比,与EGF呈反比;雄激素治疗中,睾丸重量、EGF均上升,且大于正常(P<0.01),RT骤升;停药时,EGF降至正常;雌激素治疗时,EGF亦高于正常(P<0.01),KD微上升,停药时,EGF迅速上升呈高于正常(P<0.01),睾丸重量下降。研究表明:睾丸发育不同时期、性激素水平的变化均与EGF、EGFR数目,结合力有关。
An experimental study was carried out to investigate the effect of sex hormone on the epidermal growth factor(EGF)of testis and its binding with EGF receptors.Wistan rats of different age(4,13 and 40 weeks)were used.The maximum specific binding of(125I)EGF to the testicular membrane fraction was achieved after incubation at 27℃ for 70 minutes.The age of the experimental animal was positively related to the weitht of the testis and negatively to EGF.When treated with testosterone propionate,both the weight of the testis and the EGF were elevated(P<0.01)and there was a remarkable rise of RT. With cessation of the androgen.the EGF dropped to normal. When treated with diethylstilbestrol, the EGF also raised above normal(P<0.01) with KD slightly elevated. With cessation of the drug.the EGF raised rapidly while the weight of the testis dropped. The result demonstrated that during the development of the testis.changes in sex hormone level has been correlated with the EGF,the number of EGF,and its binding ability.
