摘要
肾钙素(Nephrocalcin,NC)糖蛋白是草酸钙结石形成的主要抑制物之一。我们用改良的Nakagawa法从正常人和草酸钙结石病人尿中及草酸钙结石中提取、测定并比较了它们的分子量、等电点及氨基酸组成,发现NC为一组分子量17000,等电点范围较宽的酸性糖蛋白。结石病人由于尿NC分子中缺少γ-羧基谷氨酸,导致其对一水草酸钙(COM)晶体生长和聚集的抑制活性降低,因而有可能成为结石形成的原因之一。
Nephrocalcin (NC)glycoprotein is an essential inhibitor for calcium oxalate stone formation.We isolated and purificated NC by modified Nakagawas technique from urine of normal men and from patients with recurrent calcium oxalate urolithiasis and also from calcium oxalate calculi.We determined and compared their molecular weights,isoelectric points(PI )and amino acid compositions and found that NC is a group of acid glycoprotein with molecular weights around 17000 and PI in the area of 4.5 ̄7.0.NC from patients of calcium oxalate stone had less γ-carboxyglutamic acid (GLa)and less inhibitory effect to COM crystal growth and aggregation. This phenomenon might play a role in calcium stone formation.
