摘要
从鼠的尿液中分离出肾钙素(NC),测分子量为14600,等电点(pI)为4.5~6.0,与正常人尿NC相似;应用维生素K拮抗剂香豆素可以减少鼠尿NC中的γ-羟基谷氨酸(Gla)量,对其它氨基酸影响不大;用一水草酸钙(COM)种晶技术测量其对COM结晶生长,聚集抑制率也降低。结果提示:鼠肾产生的NC,其分子Gla是NC合成后产生,通过维生素K参与谷氨酸(Glu)羧化形成,NC分子缺少Gla可减低它对COM结晶生长,聚集抑制率。
Nephrocalcin(NC) was isolated from rat urine.The molecular weight was 14600 and the isoelectric point 4.5-6,being very similar to the urinary NC in normal human being.With the use of coumarol,a vitamin K-antagonist.it was able to reduce the r-carboxyglymin(Gla) in rat urinary NC while little effect was observed on other amino acids.The decrease of inhibitory effect on COM crystalization was also verified by the adoption of COM crystalization technique.The results denoted that the biosynthesis of Gla in NC occurs posttranslationally and involves vitamin K-dependent enzymatic carbozylation of specific glutamic acid in the polypeptide chain.Vitamin K-antagonist could reduce the turnover.It was concluded that Gla deficiency in NC plays an important role in the formation of urolithiasis.
