慢性肾衰大鼠残肾血流量和氧自由基改变及其意义 被引量:22

THE BLOOD FLOW AND OXYGEN FREE RAD-ICALS OF REMNANT KIDNEY IN CHRONIC RE-NAL FAILURE RATS

摘要 用氢清除法和电子自旋共振等方法测定了5/6肾切除大鼠术30~120天残肾的有关指标,发现饲以普通饲料的Ⅱ组大鼠残肾组织皿流量和氧自由基含量显著增加,抗氧化能力显著下降,肾功能进行性恶化,残肾炎性细胞浸润和纤维化显著;饲以大剂量维生素E的Ⅲ组大鼠,残肾血流量也显著增加,但氧自由基含量无显著增加,抗氧化能力无降低,肾功能和形态损害显著轻于Ⅱ组,动物死亡率显著低于Ⅱ组。提示残肾高灌注引起的氧自由基增高在残肾的进行性毁损中起有重要作用。 ome biochernical,physical and histological obser-vations of remnant kidney in 5/6 nephrectomy chronicrenal failure rats were performed when 30~120 daysafter Operation using hydrogen gas clearance tech-nique, electronspinresonance spectroscopy, and soon.In group Ⅱ feeded with common feed,the bloodflow and oxygen free radical content of remnant kidneytissue increased markedly compared with the sham-operation control group, accomplied by decreased renalanti oxidative ability, renal function insufficiency, andleucocyte infiltration and fibrosis in rernnant kidney. Ingroup Ⅲ feeded with feed contained high vitarnin E, theblood flow of remnant kidney tissue increased as muchas group Ⅱ, but the renal free radical content showednormal, with normal renal anti-oxidative ability , andless functional and histological impairment of remnantkidney than group Ⅱ. The mortality of group Ⅲ waslower than group Ⅱ. These data suggested that en-hancement of renal oxygen free radicals caused by hy-perperfusion played an irnportant role in the impair-ment of remnant kidney.
