摘要
以不同技术路线制备P210/MMC,P210/TNP及P40/CD3三种双功能抗体(BsAb)。在11次细胞融合中,接种7484孔,获15株二次杂交瘤,其中5株分泌BsAb。将亲本杂交瘤分别诱变为HGPRT^-及TK^-,融合后以HAT选择培养并辅以饲养细胞较易获得二次杂交瘤。证明两种抗原交互反应的桥联试验,特别是当二次杂交瘤分泌不同类别的Ig时,对于确证BsAb的存在有着重要意义。本文首次报告,γ2b与μ链间组合形成BsAb的机率为16.7%(2/12)。
Bispecific monoclonal antibodies (BsAb) P210/MMC P210/TNP and P40/CD3 were prepared with different technical approaches. Among 11 fusions, 7484 wells were planted, and 15 hybrid-hybridomas were obtained, from which only 5 secreted BsAb. It was more successful to get hybrid-hybridomas,when the parental hybridomas were induced to HGPRT- and TK- mutants for fusion and feed cells were used in HAT selective cultures. In addition,it was demonstrated that bridge assay was significant to identify the characters of BsAb,especially when hybrid-hybridomas secreted different classes of Ig. In present study,it was reported that the frequency of BsAb recombined from r2b and u was about 16. 7% (2/12).
