乙肝病毒(Adr)全基因组在转基因小鼠体内(HBV)DNA的存在状态 被引量:6

摘要 本文用PCR,SouthernBlot方法检测了34只Go代26只转基因小鼠携带HBVDNA(阳性率为76%),515只G1代转基因小鼠中248只组织中携带HBVDNA(阳性率为48%),25只G2代转基因小鼠中18只组织中携带HBVDNA(阳性率为72%).对108只尼组织阳性小鼠血清检测,102只小鼠血清呈HHVDNA阳性。以上结果证明,HBVDNA基因已整合小鼠的染色体中。这些转基因小鼠对研究HBV病毒的复制与表达,慢性肝炎以及肝癌发生的机理,治疗乙肝药物的筛选等具有十分重要的价值。 Hepatitis B virus (HBV) has an extremely narrow host range, and infects and replicates only in human and chimpanzee. The present experiment produced transgenic mice by microinjecting a partial tandem dupli-complete HBV genome adr type into fertilized ova of 34 mice,26 off springs (G0) were producers for HBV (76% HBV positive rate). The HBV genomes were transmitted to the next generation (G1) , and these G1 mice also produced HBV (248/515, 48% positive rate), 18/25 inbred G2 tail tissues carried HBV DNA (72% positive rate). Virus genome DNA was presentin sera of 102/108 G2 mice. Transgenic mouse containing HBV sequences is an useful tool for detailed study of replication and expression of HBV and for pathological study of hepatitis and hepato-cellular carcinoma.
