镍与铜联合作用致SHE细胞转化的实验研究 被引量:2

摘要 镍在国民经济中占有重要地位。镍冶炼工肺癌高发国外已有许多研究报道,我国镍精炼工肺癌死亡超量也已经证实。在镍冶炼生产中,有多种污染物共存于车间空气中,如铜、钴、多环芳烃等。它们对镍工肺癌发病的作用如何,是值得深入研究的课题。据此,我们进行了硫酸铜诱发细胞形态学转化作用、铜与镍联合作用下致细胞形态学转化作用的实验研究,为探讨铜在镍致肺癌中的作用提供科学资料。结果,硫酸铜本身没有致SHE细胞发生形态学转化,硫酸铜与硫酸镍同时或先后染毒时既没有与镍有致形态转化的协同作用,也没有促进镍所启动的SHE细胞形态学转化过程。认为,铜没有起促癌物的作用。本实验研究结果,对于铜在镍致肺癌中可能起某种作用的流行病学迹象没有给于支持。 Nickel is an important metal in industry. Excess risk of mortality from lung cancer for nickel smeltery workers has been reported abroad and demonstrated in China. However, it does not mean that such an excessive risk will occur whatever chemical form (s) of nickel exposed to. Perhaps combined effects of nickel and coexisting pollutants may play certain roles in pathogenesis of lung cancer among nickel workers.Morphological transformation of copper and combined effects of nickel and copper were purposely studied. Morphological transformation assay is an useful test in screening the carcinogenicity of chemicals. In this study, foci system was used for the transformation assay. The target cell was SHE cells, which was Prepared with monolayer culture technique. The test chemicals are nickel sulfate and copper sulfate. 3-MCA (3-Methyl-Cholanthrene) was used as a positive material. The transformation effects of CuSO4 and the combined effects of nickel and copper have not been reported before. Present results showed that nickel sulfate gives no enhancement of the transformation frequency in combination with different concentrations of copper sulfate. Copper sulfate neither induced transformation itself nor enhanced the transformation frequency in the presence of nickel.
