缺氧时肺动脉内皮细胞合成因子Ⅷ相关抗原和前列环素的变化 被引量:1

Changes of Synthesis of ⅧR: Ag and PGI_2 by Cultured Pulmonary Arterial Endothelial Cells during Hypoxia

摘要 为研究缺氧对肺动脉内皮细胞(PAEC)的影响,利用猪PAEC体外培养的方法,观察了缺氧时PAECⅧR:Ag阳性率、阳性PAEC荧光强度以及培养液中PGI_2含量的变化.结果表明,缺氧过程中,PAEC的ⅧR:Ag阳性率比对照组有明显降低,而阳性细胞的ⅧR:Ag荧光强度却比对照组明显增加.PAEC合成和分泌PGI_2的水平在缺氧开始时有所增加,但随着缺氧时间的延长却比对照组明显降低.PAEC在缺氧过程中的这些变化可能在缺氧性肺动脉高压和肺水肿的发生中发挥一定作用. In this study, the effects of hypoxia on pulmonary arterial endothelial cells (PAEC) were investigated by using cultured pig's PAEC as a model. The changes of the rates of PAEC with positive immunofluorescence for factor Ⅷ related antigen ⅧR:Ag) ,the mean intensity of immunofluorescence in positive PAEC and the concentration of prostacylin (PGI2 ) in cultured media released by PAEC during hypoxia were observed. The hypoxia exposed PAEC demonstrated a decreased rate of positive im-munofluorescensce for ⅧR: Ag as compared with control, while an increased mean intensity of positive immunofluorscence was found. Concentrations of PGI2 in the media showed an initial increase and followed by a significant decrease as compared with control during the resting period of hypoxia. These results indicated that PAEC might play an important role in hypoxic pulmonary arterial hypertension and hypoxic pulmonary edema.
