摘要
采用聚丙烯酰胺凝胶电泳方法对柑桔几个杂交组合材料进行了同工酶分析。结果表明,利用谷草转氨酶(GOT)同工酶可以鉴别华农本地早×三湖化红的杂种植株。谷草转氨酶、过氧化物酶(POX)、苹果酸脱氢酶(MDH)三种同工酶系统以及叶片形状四个指标均能充分鉴别华农本地早×宜昌橙的杂种。对华农本地早×宜昌橙F_2代进行GOT、POX同工酶遗传分析表明,GOT-2的分离符合二个或三个基因位点控制的假说;而POX-B分离广泛,尚难以进行遗传解释。对GOT-2、POX-B两种同工酶谱之间及其与叶片形状、嫩梢颜色两个形态学性状之间进行了独立测验,表明它们之间无连锁关系。
The isozymes of the progenies of several citrus hybrid combinations were analysed by polyacryamide gel electrophoresis. Following results were obtained. 5 out of 43 seedlings of Huanong Jimikan ( C.reticulata Blanco ) × Huahong ( a natural hybrid ) were identified as hybrids by GOT isozymes; 8 hybrids could be identified from 15 seed-lings of Huanong Jimikan ×Ichang papeda ( C. ichangensisi Swingle ) by anyone of the following four markers. GOT, POX, MDH isozymes and leaf shape genetic analysis were conducted with the F1, F2 proge-nies of Huanong Jimikan ×Ichang papeda, which revealed that the segregation of GOT-2 fitted the pattern of two or three gene loci; and the segregation of POX-B was too complicated to explain. Linkage studies showed that there were no linkage relationships between GOT-2 and POX-B and between the two isozymes, leaf shape and young shoot color.
