天然和氧化修饰型脂蛋白对培养人动脉平滑肌细胞形态的影响 被引量:17

Effect of Native and Oxidization-Modified LDL,VLDL and HDL on the Morphological Appearance and Phenotype of Cultured Human Arterial Smooth Muscle Cells

摘要 在建立人主动脉平滑肌细胞(SMC)体外培养方法的基础上,观察了各种天然和氧化修饰型脂蛋白对培养人SMC形态以及表型的影响。在氧化修饰低密度脂蛋白(OX-LDL),氧化修饰高密度脂蛋白(Ox-HDL)和氧化修饰极低密度脂蛋白(OX-VLDL)的作用下,SMC形态发生以下变化:胞体变大,由梭形变为不规则多边形,突起细而长,而且“谷”、“峰”样生长方式也发生改变;LDL和vLDL对SMC的形态亦有影响,但不如氧化修饰脂蛋白明显;HDL则无影响。电镜下观察,在OX-LDL的诱导下,SMC胞浆中的肌丝较对照组明显减少,而高氏器,内质网和线粒体则明显增多。上述结果说明,LDL、VLDL、OX-LDL、OX-VLDL和OX-HDL的致动脉粥样硬化作用可能与刺激SMC表型由“收缩”型向“合成”型转化有关。 The arterial smooth muscle cells(SMC) are the prdominant type of cells in atheroscle-rotic lesions, and their proliferation plays an importantrole in the process of AS genesis,We estabiished theprimary culture and sub-culture method for human ar-terial SMC and observed the effects LDL,VLDL,HDL,OX-LDL,OX-VLDL and OX-HDL on the mor-phological appearance and phenotype of cultured hu-man arterial SMC.The results revealed that the cellscultured in the medium containing OX-LDL,OX-VLDL and OX-HDL(25μg/ml ) became more irregularwith long finger-like projects and lacked a bipolarshape,and the characteristic.“hills” and “valleys” dis-appeared. LDL and vLDL also had the influence butin smaller degrees. HDL did not have any effect.TheSMC cultured in the presence of OX-LDL (25μg/ml )had less myofilaments and more endoplasmic reticu-lua, Golgi and mitochondria in cytoplasm comparedwith the cells in control group under electronmi-croscope. These results suggest that the atherogenicrole of LDL ,OX-LDL,OX-HDL,and OX-VLDL are closely related to their stimulating effectson the modulation of phenotype of arterial SMCs from“contractile”towards“synthetic”.
