2,6-吡啶二羧酸钒(Ⅲ,Ⅳ,Ⅴ)配合物透过MDCK细胞单层的能力以及与Caco-2细胞单层的比较 被引量:7

摘要 应用MDCK细胞模型研究了有降糖作用的2,6-吡啶二羧酸钒(Ⅲ)、钒(Ⅳ)、钒(V)配合物吸收能力和细胞毒性,并和Caco-2细胞模型的一些结果进行了比较.用MDCK细胞模型,实验测得3种钒配合物的表观渗透系数(Papp)分别为7.5±1.0(V(V)-dipic),1.0±0.2(V(IV)-dipic),1.7±0.4(V(Ⅲ)-dipic)(×10^-6cm/s);其中V(V)-dipic络合物有明显较大的吸收能力,这和其动物实验中的降糖效果一致.用Caco-2细胞模型测定得到相近的跨膜渗透能力(Papp=1~3×10^-6cm/s);但显著不同的是,在过量吡啶二羧酸配体存在时,3种配合物从AP→BL方向的跨MDCK细胞单层的渗透能力有较大的提高,显示在MDCK上可能存在氧钒配阴离子的主动运输方式.3种配合物对MDCK细胞的毒性相似,IC50为0.6~0.9mmol/L,略高于对Caco-2细胞的毒性(IC50=1.6~2mmol/L);其结果与渗透能力相呼应,表明钒化合物的吸收、代谢和毒性有着密切的相关关系.
