预制微血管样结构凝胶的植入研究 被引量:8

摘要 目的观察含有微血管样结构的凝胶块植入体内后与体内血液循环的连通情况,以及血管内皮细胞生长因子(VEGF165)对凝胶血管化的影响。方法在纤维蛋白凝胶中以VEGF165诱导小鼠微血管内皮细胞(MVEC)形成微血管样结构,设无VEGF165为对照,将制作的凝胶块用有微孔的塑料薄膜包裹后植入小鼠肠系膜间,2周后取样进行组织学切片,苏木素伊红(HE)染色和免疫组织化学染色,图像分析并测量血管化面积。结果凝胶植入2周见实验组凝胶周围有丰富的新生血管网经过塑料薄膜的小孔与凝胶内血管相连,对照组表面新生血管较少。组织学切片(HE染色)见VEGF组血管化总面积约为对照的50倍(P<0.01)。实验组新生血管直径10~100μm,分布于凝胶中心和周边部位;而对照组新生血管最大直径<20μm,主要分布于凝胶的周边。结论含微血管样结构的预制凝胶植入后能形成具有血流灌注的微血管网。纤维蛋白凝胶在此过程中充当了一种较为合适的细胞外基质的作用,而VEGF在这一过程中起到一种诱导、刺激和强化作用。 Objective To study the vascularization in fibrin-gels of microvessel-like structure after their implantation, and the effect of VEGF165 on this vascularization. Methods Microvessel endothelial cells (MVECs) of mouse were induced to form microvessel-like structure in fibrin-gels by VEGF165, then the fibrin-gels was implanted at mesentery of female mouse. After 2 weeks, implanted fibrin-gels were taken out for HE and immunohistochemical staining, and their vessel area was measured by image analysis. Results The area of vascularization of gel implant in VEGF group was significantly larger than that in the control group, and the microvessel-like structure in gel was connected with intracorporeal blood circulation. Conclusion The transplant of fibrin-gels of microvessel-like structure can set up blood circulation with surrounding tissues, and VEGF plays an important role in this process.
