STZ-CFA加饲高脂诱导的糖尿病大鼠模型及其眼病特点 被引量:5

摘要 目的制备糖尿病动物模型,并观察其眼病特点。方法完全弗氏佐剂(CFA)与小剂量链脲佐菌素(steptozotocin,STZ)腹腔注射,加饲高脂饲料,诱发糖尿病眼病。结果模型大鼠血糖、血甘油三酯及胆固醇升高,80%出现白内障,内皮细胞与周细胞比值和视网膜血管密度增加,晶状体电镜示典型白内障改变,视网膜及晶状体中终末糖基化产物含量较正常升高明显。结论CFA与小剂量STZ腹腔注射联用高脂饮食可成功制备糖尿病模型,并具有典型糖尿病眼病特点,是研究糖尿病及其慢性并发症的理想动物模型。其眼部病变的发生与终末糖基化产物有一定关系。 Objective To investigate the ophthalmoapathy character in diabetic rat model. Methods In order to establish rat model with hyperglycemia, Wistar rats were injected with snmll dose of steptozotocin (STZ)-CFA, and were fed by high fat diet. Results Compared with control group, postprandial blood glucose level in model rats was elevated significantly, as well as triceridemia and cholesterolemia. Eighty percent of model group were developing in cataract. The ratio of EC/PC and the density of microvascular in retina were increased greatly in these rats. And their typical cataract performance can be confirmed by electronic microscopy. It was also found that content of AGE in lens and retina were much higher than normals. Conclusion The diabetes rat model can be successfully prepared by injected with small dose of CFA and STZ, supplemented with high fat feed. This model which can imitate nature diabetes developing course, could provide a particularly advantageous tool for investigations of diabetic complication and pharmacology. The ophthalmopathy of model diabetic rats may be have some relation to AGE.
