摘要
目的:寻求不同温度下脂肪组织冻存时的适宜低温保护剂。方法:在-16℃(普通家用冻箱)、-80℃(深低温冰箱)及-196℃(液氮)条件下,加入不同的低温保护剂,冷冻脂肪组织,24h后测定肌酸激酶活力,并与贮存前酶活力做比较。结果:液氮组以15%二甲基亚砜,6%丙二醇为低温保护剂时脂肪组织活力最高,达贮存前的76%,深低温冰箱组以15%二甲基亚砜组活力最高,达66%,而普通冰箱组最高也仅为46%。结论:低温保护剂能明显保持冷冻脂肪组织活力,不同冻存条件下其最适宜的低温保护剂各异。
Objective To explore an appropriate cryoprotector for cryopreservation of adipose tissue. Methods Treated with different cryoprotectors,fat granule was stored at -16℃(domestic refrigerator),-80℃(ultra-low-temperature refrigerator)and -196℃(liquid nitrogen)respectively.Activity of creative kinase was measured 24 hours later,and contrasted with that of fresh adipose tissue. Results At -196℃,while cryoprotectors was 15% dimethyl sulphoxide and 6% propanediol,the viability of adipose tissue was the best,which reached to 76%.At -80℃,the highest viability of adipose tissue was 66% with the use of 15% dimethyl sulphoxide.However,at -16℃,the best result was only 46%. Conclusion The appropriate cryoprotector could obviously keep the viability of frozen fat,it should be chosen according to the different frozen condition.
