反义及正义α-SMA的表达对成纤维细胞与平滑肌细胞的影响 被引量:1

摘要 目的:利用α-平滑肌肌动蛋白(α-SMA)基因,重组反义和正义逆转录病毒载体,评价α-SMA逆转录病毒对NIH3T3细胞和血管平滑肌的影响。方法:用含全长α-平滑肌肌动蛋白(α-SMA)cDNA的质粒DNA的酶切、连接,重组反义和正义α-SMA的目的cDNA,构建逆转录病毒载体,并感染培养的NIH3T3细胞与平滑肌细胞,进行细胞形态学和基因表达的观察。结果:反义和正义重组逆转录病毒能够携带的目的基因并成功整合到NIH3T3细胞基因组中;利用两种逆转录病毒分别感染NIH3T3、血管平滑肌细胞,发现对细胞生长和形态有一定的影响,反义重组逆转录病毒使细胞活性下降、基因表达减弱,而正义重组逆转录病毒使细胞活性下降、基因表达增强明显。结论:α-SMA重组病毒对细胞的形态、突起的生长甚至细胞的生存均有影响。 Objective To investigate the effect ofa-smooth muscle actin (α-SMA)gene expression in NIH3T3 and smooth muscle cells,which were transfected with recombinant sense and antisense retrovirus. Methods A recombined sense and antisense α-SMA cDNA was obtained by gene recombination to clone the α-SMA cDNA to vector, then transfected into NIH3T3 and smooth muscle cells.The cells morphology and gene expression of sense and antisense α-SMA in them were observed. Results Recombined sense and antisense α-SMA cDNA carried by retrovirus can integrate into NIH3T3 cell correctly.Using both of Recombined retrovirus transfected NIH3T3 and smooth muscle cells,it was observed that their cell morphology and growth were changed. Conclusion Recombined sense and antisense α-SMA retrovirus can affect the morphology and growth ratio of transfected cells.
