摘要
目的观察海带多糖L01对血管内皮损伤老龄动物血小板活性的影响,探讨其抗血栓的作用机制。方法将老龄大鼠分为4组,分别为正常组、模型组和海带多糖L01高、低剂量组。采用注射肾上腺素法制备血管内皮损伤大鼠模型,比较过滤前后血小板计数测定大鼠血小板粘附性,火棉胶玻片法观察血小板粘附聚集活性,酶联免疫吸附双抗体夹心(EUSA)法测定血浆血管性假血友病因子(vonWillebrandFactor,vWF)和血小板α。颗粒膜糖蛋白140(GMP-140)水平。结果与模型组比较,海带多糖L01高剂量组(100mg/kg)和低剂量组(10mg/kg)均能降低血管内皮损伤老龄大鼠血小板粘附率和表面粘附聚集活性,降低血浆vWF和GMP-140水平。结论海带多糖抗血栓作用与其降低血浆vWF和GMP-140水平、抑制血小板粘附聚集有关。
Objective To investigate the effect of laminaria polysaccharide L01 on activation of platelet and to study the mechanism of laminaria polysacchadde L01 inhibiting thrombus formation. Methods The aged rats were divided into 4 groups: normal group, model group, laminaria polysaccharide L01 high-dose and low-dose group. A rat model of vascular endothelial injury was established via in}ecting adrenaline. The activation of platelet adhesion was evaluated by comparing the difference of the platelet count before and after the blood filtration. The activation of platelet adhesion-aggregation on facet was observed by collodion-glass plate method. The plasma contents of yon Willebrand Factor(vWF) and platelet a branule membrane-glycoprotein 140(GMP-140)were measured by ELISA. Results Compared with model group, the percentage of platelet adhesion, the activation of platelet adhesion-aggregation on facet and the plasme level of vWF and GMP-140 were obviously decreased by both high dose group(100mg/kg)and low group(10mg/kg)of laminaria polysaccharide L01 in aged rats of vascular endothelium in injury. Conclusion The effect of inhibiting thrombus formation of laminaria polysaccharide L01 associated with its effects of decreasing the plasma level of vWF and GMP-140 and inhibiting platelet adhesion and aggregation.
