摘要
目的:检测雄黄诱导白血病细胞凋亡的能力,并探讨其分子机制。方法:应用形态学观察检测细胞凋亡,应用检测流式细胞仪白血病细胞bcl-2和Fas蛋白的表达。结果:雄黄可诱导白血病细胞发生典型的凋亡,白血病细胞有bcl-2强表达,同时雄黄可使白血病细胞bcl-2蛋白表达呈时间依赖性的下降。白血病细胞有较弱的Fas蛋白表达,随着雄黄与白血病细胞作用时间的延长,未见Fas蛋白表达有明显改变。结论:雄黄通过影响白血病细胞bcl-2蛋白的表达从而导致白血病细胞发生凋亡是其治疗白血病的重要分子机制。
Objective To investigate the apoptosis of leukemic cells induced by realgar,and illustrate the possibe molecular mechanism. Methods Apoptosis was detected by morphological observation, bcl-2 and Fas expression was detected by flow cytometric analysis. Results There was strong expression of bcl-2 in leukemic cells. Realgar can induce leukemic cells to apoptosis and down-regulated bcl 2 expression. Fas protein expression in leukemic cells was very low ,realgar induced apoptosis of leukemic cells ,but didn't induce Fas protein expression. Conclusion Realgar can induce apoptosis in leukemic cells. Down regulation of bcl-2 expression appears to play animportant role in the apoptosis of the differentiated leukemic cells.
