反相高效液相色谱法测定盐酸青藤碱纳米脂质体药物含量及包封率 被引量:13

摘要 目的建立盐酸青藤碱纳米脂质体药物含量及包封率的测定方法。方法采用反相高效液相色谱法,以十八烷基硅烷键合硅胶为固定相,以甲醇0.01mol·L-1NaH2PO4(38∶62)为流动相;柱温为30℃;流速为1mL·min-1;检测波长为262nm。采用透析法分离盐酸青藤碱游离药物。结果在所选定的色谱条件下辅料及溶剂对盐酸青藤碱的测定无干扰,盐酸青藤碱在5~80μg·mL-1线性关系良好(r=0.9999);回收率为99.6%~101.1%,日内和日间RSD均<2%(n=5)。结论该方法准确可靠、简单快速,可用于盐酸青藤碱纳米脂质体含量及包封率的测定。 OBJECTIVE To establish a method for the content and entrapment efficiency determination of sinomenine hydrochloride nanoliposome. METHODS The content of sinomenine hydrochloride was determined by RP-HPLC and the conditions were as follows: a packed C18 column, a mobile phase consisting of methanol-0.01mol·L^-1NaH2PO4 (38:62), a flow rate of 1 mL·min^-1, a detection wavelength of 262 nm and the temperature was 30℃.The free sinomenine hydrochloride was separated by dialysis. RESULTS Sinomenine hydrochloride had a good linear relation in the range of 5-80μg·mL^-1 (r=0. 999 9), the intra day and inter-day RSD were less than 2%, and the average recovery was between 99.6-101.1%. CONCLUSIONS A simple, rapid, and accurate method for the determination of sinomenine hydrochloride nanoliposome and their entrapment efficiencv is established.
