摘要
目的:探讨约氏疟原虫感染早期小鼠免疫应答的差异性。方法:约氏疟原虫腹腔感染DBA/2、BALB/c和C57BL/6小鼠,制备薄血膜,姬姆萨染色,计数红细胞感染率;通过ELISA和Griess实验分别检测脾细胞培养上清中γ干扰素(IFN-γ)和一氧化氮(NO)水平。结果:感染第3d之后,DBA/2小鼠的虫体血症水平显著低于BALB/c和C57BL/6小鼠;将疟原虫寄生的红细胞(PRBC)和脂多糖(LPS)或IFN-γ分别与3种感染第3d小鼠脾细胞共同培养后,DBA/2小鼠脾细胞培养上清中的IFN-γ和NO水平明显高于BALB/c和C57BL/6小鼠。结论:约氏疟原虫感染早期,不同遗传背景小鼠在Th1型细胞免疫应答的建立和巨噬细胞活化方面存在显著差异。
Objective: To investigate the difference in immune response to Plasmodium yoelii ( P. yoelii ) infection in 3 kinds of mice. Methods: DBA/2 ,BALB/c and C57BL/6 mice were infected with P. yoelii intraperitoneal injection of red blood cells with P. yoelii parasitized. Parasitemia of mice was monitored by observing microscopically the blood smears stained with Giemsa. The levels of nitric oxide and interferon-γ(IFN-γ) in spleen cell cultured supcrnatants of 3 kinds of infected mice were measured by using ELISA kit and Griess reaction. Results: On the 3rd day after infection, the parasitemia of DBA/2 mice was much lower than that of BALB/c and C57BL/6 mice. The levels of nitric oxide and IFN-γ in spleen cells of DBA/2 mice were much higher than those of BALN/c and C57BL/6 mice when the spleen cells of 3 kinds of mice were cultivated with different stimuli. Conclusion: There exist remarkable differences in the Th1 immune response to P. yoelii infection and the activation of macrophage in different kinds of mice。
