纳洛酮对缺氧缺血新生大鼠视网膜损伤线粒体Ca^(2+)质量分数的影响

摘要 目的观察纳洛酮对新生大鼠缺氧缺血损伤后视网膜线粒体Ca2+质量分数的影响,探讨纳洛酮对缺氧缺血视网膜损伤的保护作用。方法新生健康SD大鼠36只,随机分为对照、缺氧缺血、纳洛酮保护3组,每组12只。将新生SD大鼠结扎右颈总动脉并吸入96%N2+4%O2制成缺氧缺血模型,以原子吸收火焰法测定线粒体Ca2+质量分数。结果对照组、缺氧缺血组、纳洛酮保护组Ca2+质量分数(μg/gpro)分别为31·2±0·03,98·5±3·49和40·3±1·61。结论纳洛酮可有效降低缺氧缺血视网膜损伤后线粒体Ca2+质量分数,从而减轻缺氧缺血对视网膜的损伤。 Objective To observe the mitochondrial calcium level changes in hypoxic and ischemic retina and study the protection to retina from naloxone damage. Methods Thirty-six newly born rats were divided into control group, hypoxic and ischimic group and naloxone protection group,and 12 rats were included for each group. The right carotidartery were ligated and animal model was created by exposure the rats to 96% N2 and 4% O2 in SD rats. Mitochondrial calcium concentration in retina of the rats was measured with atomic absorption spectrophotometry. Results Mitochondrial calcium concentrations in retina of newly born rats were 31.2±0. 03,98.5±3.49 and 40.3±1. 61μg/g protein in control group, hypoxic and ischimic group and naloxone treatment group, respectively, and increased mitochondrial calcium concentration in retina under the hypoxic and ischimic status was found when compared to the control group( P 〈 0. 01 ). The mitochondrial calcium concentration in retina was statistically decreased in naloxone treated group in comparison with the hypoxic and ischimic group (P 〈 0.01 ) and was insignificantly higher than the control group ( P 〉 0. 05 ). Conclusion Naloxone can effectively attenuate retinal damage induced by hypoxia and ischemia by reducing retinal mitochondrial calcium levels in newly born rats.
