摘要
目的:观察大蒜油对HL-60细胞的诱导分化作用。方法:MTT法检测大蒜油对HL-60细胞的增殖抑制,Giemsa染色观察细胞形态改变,流式细胞术分析细胞周期改变,并检测CD11b、CD15的表达。RT-PCR检测bcl-2、c-myc/mad、mpo基因mRNA的表达。同时设ATRA、As2O3为阳性对照,与大蒜油的作用进行比较分析。结果:大蒜油明显抑制了HL-60细胞增殖,使大部分细胞阻滞于G0/G1期,作用强于ATRA和As2O3;诱导细胞成熟分化的能力与ATRA接近,优于As2O3;通过抑制bcl-2、c-myc和mpo基因的表达,促进mad基因的表达来发挥作用。
Objective: To study the effect of garlic oil on inducing HL-60 cell differentiation. Methods: The inhibition rates of cellular proliferation were measured by MTT. The morphology changes were observed after cells were stained by Giemsa. Cell cycle distribution and expressions of surface antigen CD11b, CD15 were analyzed by flow cytometry. The expressions of bcl-2, c-myc/mad, mpo genes were determined by RT-PCR. Meanwhile, ATRA and As2O3 were usedeffects in inhibiting HL-60 cell proliferation thanas positive controls. Results: The garlic oil had strongerATRA and As2O3. The cell cycle was blocked in G0/ G1 phase. The garlic oil also had a similar effect in promoting HL-60 cell differentiation as that of ATRA. The mechanism may be due to its decreasing bcl-2, c-myc, and mpo gene expressions and increasing of mad gene expression. Conclusions: Garlic oil can significantly inhibit HL-60 cell proliferation and induce differentiation. It may become a new agent in leukemia treatment.
