摘要
用含有不同浓度NaCl的MS诱导培养基,采用直接筛选和逐级筛选法,对结缕草Zenith品种进行耐盐变异体的选择研究。结果表明:NaCl浓度对Zenith品种愈伤诱导率的影响极显著(p<0.01),在NaCl浓度为1.0%时已很难诱导出愈伤组织;愈伤组织直接在含有不同浓度NaCl培养基上选择表明NaCl浓度对愈伤组织相对生长量影响极显著(p<0.01);通过逐步筛选,愈伤组织的存活率显著增加;两种筛选方法获得的耐盐愈伤组织再生后分别获得耐盐1.0%及1.2%的植株。
Zoysiagrass cv.Zenith was used to select salt-tolerant variants on MS medium containing different concentration of NaC1 via direct selection and continuous selection. The result showed that NaC1 had remarkable influence on callus inducement, it was very difficult to induce callus when the concentration of NaC1 increase to 1.0%. The concentration of NaC1 also have great influence on relative growth rate when callus selected on medium containing different concentration of NaC1. The survival rate of callus increased after selection step by step. Regenerate plant derived from direct selection could tolerate up to 1.0% NaC1, while continuous selection could tolerate up to 1.2% NaC1.
