摘要
目的:观察和探讨灵芝孢子粉对癫痫大鼠脑组织细胞因子IL-6含量变化的影响。方法:10周龄Wistar雄性大鼠30只,体重(150±10)g,随机分成3组,每组10只,分别是灵芝孢子粉组、癫痫模型组和空白对照组。灵芝孢子粉组和癫痫模型组用戊四氮(PTZ)亚惊厥剂量(32mg/kg),每日1次,腹腔注射28d,停药1周,再用相同剂量的PTZ测试,凡显示连续5次2级以上惊厥的大鼠为达到点燃标准,空白对照组用同体积生理盐水腹腔注射。同时灵芝孢子粉组经灌胃给予灵芝孢子粉(灌胃前禁食6h),灌胃体积为1.0ml/次;癫痫模型组和空白对照组经灌胃给予同等体积的生理盐水。实验结束处死动物,在低温条件下迅速取出鼠脑,用放射免疫法测定脑组织细胞因子IL-6含量变化。结果:灵芝孢子粉组与癫痫模型组动物相比脑组织IL-6明显降低(P<0.05)。结论:灵芝孢子粉能够有效降低癫痫大鼠脑组织细胞因子IL-6水平,纠正免疫失调而起到抗癫痫作用。
Objective.. To observe and discuss the influence of Garoderma lucidum spores on the cell factor IL--6 in the brain of epilepsy mouse. Methods: Thirty Wistar male mice at the age of 10 weeks : (150±10)g were divided into 3 groups at random(10 mice for each group): Garoderma lucidum spores group , epilepsy model group and blank contrast group. The sub--ec|ampsia dosage (32mg/kg) of Pentetrazole(PTZ) was used in Garoderma lucidum spores group and epilepsy model group, once a day by abdominal cavity injection for 28 days, and one week after stopping injection, the test was done with PTZ of the same dosage, and the mouse that showing eclampsia of more than 2 grades 5 times in succession reached the "igniting" standard, use physiological saline of the same volume was used in blank contrast group by abdominal cavity injection. Garoderma lucidum spores group is given Garoderma lucidum spores to the stomach (prohibit to eat 6h before affusing) at the same time, the allusion volume to the stomach was 1. 0ml / time ; Epilepsy model group and blank contrast group were given physiological saline of the same volume. When Finished, putting The animals were killed when the experiment was finished, and mouse's brains were taken out rapidly in the low--temperature condition , and the change of IL--6 was measured by means of RIA. Result: We compared the Garoderma lucidum spores group with the epilepsy model group, the IL-- 6 in brain tissue reduced obviously ( P 〈 0. 05 ). Conclusion: Garoderma lucidum spores can effectively reduce the cell factor IL--6 in the brain of epilepsy mouse, correct the immunity dysfunction and play a role in anti--epilepsy.
