摘要
【目的】探讨米非司酮对子宫肌瘤组织增殖细胞核抗原(PCNA)、微血管密度(MVD)的影响及其相互之间是否具相关性。【方法】40例需手术治疗(子宫全切术、子宫次全切除术、肌瘤剔除术)子宫肌瘤患者随机分为对照组与观察组。观察组术前每日口服米非司酮10 mg,连服3个月;对照组术前3月未服任何药物。用免疫组化SP法测定子宫肌瘤组织中PCNA、MVD的表达。【结果】观察组子宫肌瘤组织中PCNA、微血管密度计数表达强度均低于对照组,且两者之间存在正相关。【结论】米非司酮可能通过抑制子宫肌瘤组织中PCNA、MVD的表达降低,使肿瘤细胞生长受抑而瘤体缩小。
[Objective]To study the effects of mifepristone on the expression of proliferating cell nuclear antigen(PCNA) and microvessel density(MVD) in human uterine leiomyoma cells and investigate the relationship between them. [Methods]Forty patients with uterine leiomyoma for surgery were divided into two groups. One was control group who underwent surgery without any medicine pretreatment; the other was mifepristone group who underwent surgery after pretreatment with mifepristone 10 mg daily for 3months. After surgery, the expression of PCNA and MVD in leiomyoma was investigated by immunohistochemical techniques. [Results] Among 40 patients with leiomyoma for surgery, the expression of PCNA in mifepristone group[( 17. 885 ± 6.79)%] was lower than the control group[(31. 195± 10. 10)%]( P 〈0. 001). The microvessel density in mifepristone group[(26. 417±5.64) %] was lower than the control group[(49. 6665±8.25) %]( P 〈0. 001). The positive correlation was found between PCNA and MVD (r = 0. 927, P 〈 0. 001) in leiomyoma. [Conclusion]These findings suggest that:①Mifepristone inhibits the proliferation of leiomyoma cells. ②Mifepriston decreases MVD in leiomyoma and inhibits angiogenesis.③Mifepristone inhibits the expression of PCNA leading to the low expression of MVD. This may be the regression mechanism of mifepristone in treating uterine leiomyoma.
