摘要
本文报道用镀铜镉还原法测定血清硝酸盐。在甘氨酸-氢氧化钠缓冲液中(pH9.7),用镀有铜的镉颗粒还原血清中的硝酸盐为亚硝酸盐。亚硝酸盐与磺胺进行重氮化反应,并与N-1-萘乙二胺盐酸盐偶联。偶联产物在545nm处有吸收峰。还原最大值(75%)出现在反应1小时左右。亚硝酸盐产量正比于硝酸盐浓度至250μmol/L。检测限为1.9μmol/L,批内变异系数(CV)为7.4%,平均回收率为109.1%。16例健康人测得结果为73.1±16.1(x±s)μmol/L。由于采用标准加入法,消除了血清中的抑制物干扰。
The serum nitrate are measured with a copper-coated cadmium reduction method. After serum samples were deproteinized with Somogyi reagent.the indicated amount of nitrate standard were added to deproteinized samples.The nitrate was reduced by Cu-coated Cd granules in glycine-NaOH buffer (pHg. 7) . The nitrite produced was determined by diazotization of sulfanilamide and coupling to N-(1-naphthyl) ethylenediamine dihydrochloride.After 20 min. read adsorbances (A) against the blank at 545 nm. The maximum reduction (75%) occurred at about 1 hour.Nitrite production was proportional to nitrate concentration up to 250 μmol/L. The detection limit was 1. 9μmol/L. The repeated assay gave CV 7. 4 % for a mixed serum (n= 9) . The mean recovery of nitrate was 109. 1%.The effect of inhibitors in samples was obviated by the use of internal standards. The mean concentration of nitrate in serum of normal volunteers was 73.1 ± 16. 1μmol/L. This method is suitable to clinical laboratories.
