禾本科植物原生质体培养的研究进展 被引量:1

摘要 近年来,生物技术中最重要的进展之一就是植物原生质体培养和融合的研究。利用这项新技术,不仅大大促进了细胞生物学、植物生理学及遗传学中许多理论问题的研究,而且为作物的品种改良和育种提供了新颖的工具和方法,取得了可喜的进展。十年来的突破性进展表现在许多重要农作物,特别是禾本科作物中,如水稻、小麦、玉米、小米、高梁等原生质体培养得到再生植株。本文将对以上系列研究的进展、原生质体培养的方法和有关研究的意义进行介绍和论述。 This paper edpounds the research progress of protoplast culture in Gramineae phots,introduces lutely reactive techniques and methods and discusses its significance in theoretical research and productive utilization.About the process of protoplast culture in Gramineaephot people should pay more attention to the following points:1'To CHOOSE A PROPER GENOTYPE Form -formative prOCess is not only determined by hereditary faCtors,but also by many physiological and environmental factors.Therefore'we should select carefully for many times to choose the proper genotype which has thestronger regeneration capacity'2' TO BUILD UP THE EMBRYONIC SUSPENSION TO GET PROTOPLAST First isto culture the vigorous callus with hash division potential,and then to choose the embryoniccelle from the vigorous suspension and transfer them into the liquid culture system to build theembryonic suspension.Thus the living protoplasts can be freed and prepared from the suspension cells 3. TO ADJUST CULTURE MEDIUM IN DIFFERENT PERIODS To choose and adjustthe culture medium is very important not only to the callus culture, but also to the suspensionculture and protoplast culture.So we should have theoretical knowledge and experience aboutit'Paint protoplast has a totipotency as well as a single cell' So we can use it to improve someproperty and quality of the important crops the to create new hybrid with high tolerance andproductivity' Besides,we can use protoplast to explore cell division mechanism,membrane'snature and function,and use it as an ideal acceptor in genetic engineering.
