SDS-PAGE鉴定霍乱弧菌不同菌株外膜蛋白成分

摘要 霍乱弧菌569B等菌株用EDTA-溶菌酶法提取其外膜蛋白,以高速离心法纯化并以聚丙烯酰胺凝胶电泳分离其主要蛋白区带。经SDS-PAGE及免疫电泳等方法鉴定,表明不同菌株的外膜蛋白均显示一主要区带,其分子量为25KDa,免疫双扩试验表明不同霍乱弧菌菌株的外膜上都有一个共同的蛋白抗原。 Vibrio cholerae,like other Gram-negative hacteria,has an outer membrane containing lipopolysaccharides(LPS)and outer membrarle proteins (OMP).Both One and LPS are essential in the making of cholera vaccine.This investigation is aimed to analyze the OMP of the vibrio cholerae,helping to get an in-depth knowledge on the chemical composition of the bacterial cell envelope in the making of cholerae vaccine,Two biotypes of vibrio cholerae-classical 569B and E Tor 35A3 and NH43 were used in this study. Isolation of OMP was accomplished by treating the cell envelope with 1% lysozyme for 2 hr.at 25℃.followed by centrifugation at 105000g for 1hr.The composition of OMP was analyzed by polyacrylamide slab gel electrophoresis and SDS-PAGE. The results demonstrated the presence of a common protein with a molecular weight of 25 KDa.
