用聚合酶链反应检测宫颈癌组织中人巨细胞病毒DNA相关序列 被引量:1

摘要 为建立检测人巨细胞病毒(HCMV)感染的PCR方法并探讨HCMV与宫颈癌的关系,从石蜡包埋和活检宫颈癌组织中提取DNA,用聚合酶链反应体外扩增HCMVEcoRID片段上一段长152bp的核酸序列,再用地高辛末端转移酶标记法标记和扩增片段互补的HCMV寡核苷酸探针,用该探针证实扩增片段的特异性。检测宫颈癌标本43例,阳性率为25.6%(11/43),正常宫颈组织8例,阳性率为0。宫颈癌标本阳性率高于正常宫颈标本,提示HCMV与宫颈癌的发生有关。 To establish the PCR method for detection of HCMV and study the relationship between HCMV and cervical cancer, the authors extracted DNA from paraffin-embedded and biopsy samples of cervical cancer, and amplified a 152bp DNA sequence in HCMV EcoRI D fragment by PCR. DIG labeled HCMV oligonucleiotide probe was used to analyse the specificity of the PCR products. As a result, 11 of 43 samples of cervical cancer were positive (25.6%), none of & samples of normal cervix was positive. The result indicated that HCMV was related to cervical cancer.
