摘要
本文首先用α-32P标记的热休克蛋白70(hsP70)cDNA为探针,研究了背部烫伤对大鼠脑肝hsp70mRNA的诱导。结果表明,烫伤大鼠脑肝hsp70mRNA的表达明显增加,烫伤后5min内开始出现,24h后恢复正常。由于烫伤后脑肝温度的增加不超过1℃。因此温度升高不大可能是该基因表达增加的原因。在此基础上用hsP70的抗体,采用Westernblot的方法,分析了烫伤大鼠脑肝hsp70的变化,结果表明脑hsp70在烫伤后5min内就开始增加,烫伤后3h仍未恢复,本研究表明在完整动物局部的烫伤可以引起远隔器官(脑肝)hsp70在mRNA和蛋白水平的迅速表达,说明在整体动物hsp基因的表达可以不依赖于应激原对细胞的直接作用。
Induction of hsp70 mRNA in rats after scalding was analysed using 32P labeled hSP70 cDNA. The results showed .that the expression of hsp70 mRNA in brain and liver was induced markedly after scalding. Time course of the increase showed that induction took place within 5 min and subsided 24 h after scalding. In the liver, induction took place still earlier, i. e. within 1 min and lasted for more than 24 h. The induction of hsp70 mRNA in rats after scalding did not seem to be due to elevation of core temperature since the increase of temperature of rat brain and liver after scalding was not more than 1℃. Western blot anlyses further showed that the induction of hsp70 occured within 5 min and lasted for more than 3 h in the brain of rats after scalding. The above results demonstrated that the hsp expression in internal organs could be induced by local injuries and was not necessarily dependent upon the direct effect of stimuli in intact animals.
