摘要
红霉素链霉菌14-74经一系列诱变筛选得到1-25菌株。本文报道1-25菌株培养特性,在摇瓶及生产罐中1-25菌株的生产能力比对照14-74菌株提高7%以上。发酵液中红霉素C的含量等于或小于对照菌株。高产的遗传性能稳定,对红霉素及丙醇的耐受力明显高于亲株,是一株优于14-74菌株的高产优质的抗反馈及抗阻遏的突变菌株。
Selection of mutants with high productivity of erythromycifl was attempted. A mutant S. erythreus No. 85-3-46 was isolated from the parent strain 14-74 by preparing its protoplast then irradiation with UV and other mutagents.
The mutant 85-3-46 was further mutated with UV light and the high producer S. erythreus 1-25 was obtained by an improved selection procedure of mutants by screening the resistant mutants to phage of S. erythreus, to erythromycin, to n-propa-nol, and to the analog of methionine.
The Yield of Erythromycin of the mutant S. erythreus 1-25 was 7% higher than the parent strain in the shaking flask culture and the tank production.
It was shown that the genetics property of the high productivity was stable.
S. erythreus 1-25 strain was obviously different from the parent strain in resistance to erythromycin and to n-propanol.
Comparing with the parent strain the content of the undesirable product erythro mycin C detected by TLC in the mutant 1-25 was the same or smaller.
The result of this study showed that S.erythreus 1-25 was a high producing strain with resistance to feed back inhibition and repression.
