摘要
本研究以姜片虫成虫纯化抗原(PAA)(蛋白含量为10μg/ml)致敏经醛化和鞣化的人“O”型血红细胞,采用间接血凝试验(IHA)检测了69份姜片虫感染者血清和60份正常人血清,若以抗体滴度≥1:10者为阳性,则两者的阳性率分别为81.2%和6.7%(P<0.005);此外,还检测了日本血吸虫、肺吸虫、华支睾吸虫和长膜壳综虫感染者血清,其交叉反应率分别为10.0%(4/40)、12.5%(1/8)、0(0/8)和0(0/4)。结果表明,该法具有较好的敏感性和特异性。
In this paper, we attempt to eValuate the effect of IHA on immunodiagnosis of fasciolopsiasis. The red blood cells were collected from a donor with blood group O, and treated with glutaraldehyde and tannic acid.The prepared erythrocytes were mixed with pure antigen from adult worms of Fasciolopsis buski(PAA) (the concentration of protein was adjusted to approximately 10 μg/ml). Antigen solution used for sensitization of red blood cells was reused up to four times without loss of sensitivity.The antigen-coated red bind cells were used to detect the antibodies in sera from patients infected with either F.buski (EPG ranged 8-1344) or other parasites by using IHA,and normal controls were included. The reaction was considered to be positive when the antibody titre was≥1:10.The positive rates of the patients infected with F. buski and healthy persons were 81.2%(56/69) and 6.7%(4/60) respectively (P<0.005). In addition,the cross-reaction rates of patients with schistosomiasis japonica, parsaonimiasis westermani,clonorchiasis sinensis and hymenolepiasis diminuta were 10.0%(4/40), 12.5%(1/8),0(0/8) and 0(0/4) respectively. It is demonstrated that the IHA test has good sensitivity and speciality for immunodingnosis of fasciolopsinsis.The prospects for applying were discussed preliminarily.
