摘要
观察了醋酸铅1,5,10,50,100μmol/L对培养的LLC-PK1细胞存活率、细胞内钾离子和细胞上清液中LDH和γ-GT的影响。结果50μmol/L和100μmol/L铅作用32小时细胞的存活率已明显减少;5μmol/L铅处理1小时,细胞上清液中LDH明显升高,并在1~10μmol/L范围内存在明显的时间-效应、剂量-效应关系;50μmol/L铅处理半小时,就出现细胞内钾离子明显减少;1~100μmol/L铅处理4小时和8小时,γ-GT已明显增加,并存在剂量-效应关系。加锌20μmol/L可使细胞的存活率增加,细胞内钾离子和γ-GT、LDH的漏出明显减少。上述结果说明,醋酸铅作用于LLC-pK1细胞,首先引起细胞膜对钾离子通透性的改变,其次为LDH,当明显损伤细胞膜时可使膜结合酶γ-GT脱落,上述指标在一定范围内存在时间-效应和剂量-效应关系。20μmol/L锌在一定程度上可保护细胞膜,拮抗铅对肾小管细胞LLC-pK1的损伤作用。
In this study ,changes of cellular vitalities ,cellular K +contents,supernatant LDH and r-GT activity of cullured LLC-PK1 cell induced by 1,5,10,50 ,100μmol/L acetate lead (Pb) were investigeted ,the results showed that the cellular vitality decreased signiffcantly after 32 hours of 50and 100μmol/L.Pb treatment;the supernatant LDH actvity increased significantly after 1 hour of 5μmol/L Pb treatment ,and showd a time-effect and dose-response relationship between 1 and 10μmol/L Pb ,after half and hor of 50 μmol/L Pb treatment ,the cellular K+ contents deceased significantly;activties of r-GT increased sigficantly after 4 and 8 hour of Pb treatment and showed a dos-response relationship.addition of 20 μmol/L sulfate Zinc (Zn) showed an effect to make the significant increases of cellular vitalities and the significant decereases of leakage of cellular K+,LDH and r-GT,these results suggest that Pb act on the menbrane of the LLC-Pkl cells and make the changes of membrane K+ premeability firstly ,followed by the changes of LDH activities ,while r-GT,a membrane combine enzyme, detachede only when the membrane is heavily damaged.the parmeters showed a time-effect and dose-response relationship among certainranges.20μmol/L Zn have some protective effect on membrane damage and against the toxicity effect of Pb to kidney tubules.
